    Most Significant Contribution to Research

    Contribution #1: Identifying the Role of Autophagy in Regulation of Cellular Response to Stress  

    I have been investigating how autophagy maintains cellular hemostasis under stress conditions to help cell survival, and have successfully addressed fundamental questions in this field. I showed that autophagy actively cross-talks with several different mechanisms that determine cell viability, including apoptosis and unfolded protein response. I have shown that autophagy is the major determinant in regulating cell viability under stress conditions and that it can regulate cell death by positive or negative regulation of apoptosis. In a recent investigation my group has showed that TiO2 nanoparticle induced reactive oxygen-induced cell death which is tightly regulated by autophagy. My group has also shown that autophagy is involved in physiological cycle of hepatitis virus B and C infection and replication in human liver biopsies and infected patients. As a post-doctoral fellow I also showed that the Bcl2 protein family can regulate cross-talk of autophagy, apoptosis, and unfolded protein response in the lung and heart mesenchymal cells, and in different epithelial cells. My investigations also showed that autophagy clears damaged cellular organelles during cellular stress, which usually prevents extreme cellular death via apoptosis and provides the best chance for cellular survival. Recently, I led international teams of expert and prepared two book chapter on the regulation of autophagy by unfolded protein response, and importance of autophagy in cerebellum development (article#8) which has been published by the Elsevier and springer publishing group. 

    Contribution #2: Regulation of lung the cellular Phenotype by Autophagy

    After establishing my independent research program, my group has focused on the mechanisms involved in regulation of changes of lung epithelial mesenchymal phenotypes. Our investigations have shown that autophagy and unfolded protein response (article #3) are essential steps in regulation of lung cellular phenotype. Based on my group recent investigations about the role of autophagy in regulation of the cellular phenotype, we have addressed proper conditions for chemical inhibition of autophagy (Bafilomycin A1) in latest autophagy monitoring guideline. 

    Fibroblasts have an important function in providing a suitable response for different organelle adaptation to environmental stress. One of these responses, fibrosis, is involved in wound healing. Fibroblast-to-myofibroblast conversion is involved in the mechanism of fibrosis. As a postdoctoral trainee I have provided novel evidence that autophagy is a regulator for fibronectin and collagen Iα2 biosynthesis in both lung and heart.

    Contribution #3: Development of Methods in Cell Biology

    Since establishment of my research group, one of my major aims of research program is development of cell biology protocols which can be widely used by researchers and trainees. My HQPs have been actively involved in developing such protocols which closely related to their research training program. We (including my MSc HQPs and PDF HQP) have been able to develop a unique sensitive method for simultaneous detection of autophagy and EMT in lung epithelial cells (article #1) and measurement of small Rho GTPase activity in different types of cells. Both of these model is modified and developed so can be done with minimal equipment with highest sensitivities. I have also developed/modified using autophagy inhibitors in the latest guideline for autophagy, which is highly appreciated in autophagy community. As a member of autophagy community, I have also been involved in 2012 guidelines of autophagy protocol which broadly address the detection methods for autophagy.      

    Contribution #4: Cholesterol Metabolism Pathway and cell death mechanisms:

    My investigations have addressed the mechanism that is involved in regulation of cholesterol metabolism inhibition induced cell death in more than 20 models of cells. In 2017, I edited of two volumes of Current Molecular Pharmacology (January, April) focusing on mevalonate cascade in health and diseases.
